Your best bet if you plan to run the whole river.

Drive to Agnew Lake Lodge
Pick up shuttle driver there
Drive to Sudbury train station (about an hour)
You finish the river back at Agnew Lake Lodge where your vehicle awaits you

The train runs north every second day so you just have to match your arrival at Agnew Lake. They can do the shuttle early enough that you can get to the train which leaves at 9:00am (check to make sure that's still true) or you can rent a cabin for the night from the Lodge.

The current owner of the lodge (as of 2016) is very canoe friendly, they also handle the permits you need for the river. They had the place up for sale last time I was there, hope whomever takes over is also canoe friendly because the owners during the 80's & 90's were definitely not (although they would be happy to take your money for parking).

If you were tight for time instead of going all the way to Biscotasing you could get the train to drop you off at Sinker Creek but if you can, doing Bisco to Agnew Lake is the best.

There are other options if you want to take out at The Elbow, would save you 3 days but you miss a lot of really nice paddling and the shuttles are a bit more expensive.
